A group out on safari are awakened by a scary, other worldly sound. As they come out of their tenants they see a horde of animals that look to be possessed. These demonic animals chase the people from their campground to the nearby river. After this opening scene, time for the gang to get involved.
The gang is headed out on their own safari, where they run into the super affectionate couple - Henry and Honey Hunsecker. The Hunseckers are glad to be on this trip telling the gang that this is their 3rd safari. On arrival, the gang and the Hunseckers are loaded on a boat for a ride down the river with Cap'n Robbie along with another visitor Lloyd Mbuku. Through his binoculars, Fred spots an animal with an eerie yellow glow over its body. Suddenly, up ahead they realize that they are heading towards a deadly waterfall and the Captain is unable to control the boat. They crash down to the bottom of the river.
The gang awake on one side of the river and quickly determined that they are separated from rest of the visitors and the Captain, who are on the other side of the river. The gang travels down the river on foot and run into a giraffe and a gorilla, but are amazed by how few animals are around. After finding a place to camp for the night, the gang is soon terrorized by the possessed animals.
After the dust settles, the gang realizes that Scooby Doo and Jacko (Mbuku's monkey) are missing. As part of their search, Daphne and Velma find Mbuku's police badge. Mbuku's monkey eventually appears, but instead of being friendly, he is now angry and vicious. Velma changes his behavior back by playing music. Once he is back to normal, the gang discovers that the "glowing" of the possessed animals is staged.
The gang eventually comes upon an abandoned ship docked on the side of the river. After investigating, the gang find Scooby and all of the other animals. All clues point to the Hunseckers and the gang, with the help of the released animals, captures them.
